Comparison

FOXO4-DRI vs LL-37

Function

While FOXO4-DRI acts as a senolytic in preclinical models by promoting apoptosis of senescent cells, improving tissue function and healthspan measures in aged animals38, LL-37 acts as a broad-spectrum antimicrobial and immunomodulatory peptide involved in host defense and wound repair, but can also promote inflammation and cancer cell proliferation in some contexts5215.

Mechanism

While FOXO4-DRI works as a D-retro-inverso peptide derived from a FOXO4 region that competes with endogenous FOXO4 for binding to p53, thereby releasing p53 to trigger apoptosis selectively in senescent cells38, LL-37 is a cationic amphipathic 37-amino-acid cathelicidin peptide generated from hCAP18 that disrupts microbial membranes and modulates innate immunity, including chemotaxis, cytokine induction, and NET formation521586.

Receptor

FOXO4-DRI

Intracellular target is the FOXO4–p53 protein–protein interaction interface; it does not signal via a traditional cell-surface receptor38

LL-37

Functions as a ligand for CXCR2 on neutrophils and other myeloid cells; LL-37 signaling has also been linked to FPR2 and P2X7 in various cell types515

Summary

FOXO4-DRI and LL-37 are noticeably different, with limited direct overlap in their usual biological context. Their typical research and application settings separate fairly clearly: FOXO4-DRI is more often discussed in the realm of Aging and longevity, whereas LL-37 is more often associated with the realm of Immunology and inflammation and Gastroenterology. Their biological logic is quite different: FOXO4-DRI is a protein interaction inhibitor, whereas LL-37 is a membrane disruptor and a signaling modulator. FOXO4-DRI has a more synthetic design origin, while LL-37 is closer to natural endogenous background and both are still best understood as being in Preclinical development. FOXO4-DRI incorporates retro-inverso isomerization features that are not part of LL-37, while their sequence patterns also diverge, with FOXO4-DRI showing protein-mimetic sequence features and LL-37 showing cationic amphipathic and alpha-helical domain features.
