Mechanism

While FOXO4-DRI works as a D-retro-inverso peptide derived from a FOXO4 region that competes with endogenous FOXO4 for binding to p53, thereby releasing p53 to trigger apoptosis selectively in senescent cells38, KPV is the C-terminal Lys-Pro-Val tripeptide fragment of α-MSH, which exerts potent anti-inflammatory effects largely via inhibition of NF-κB signaling and modulation of cytokine expression, with many actions independent of classical melanocortin receptor activation684.

FOXO4-DRI and KPV are noticeably different, with limited direct overlap in their usual biological context.
