Comparison

Epitalon vs Sermorelin

Function

While Epitalon is reported to increase lifespan and reduce tumor incidence in animals, and to influence differentiation of neural and mesenchymal stem cells; is studied as an epigenetic regulatory peptide36, Sermorelin is used as a diagnostic agent for assessing GH secretory capacity and as a research tool or off-label therapy to increase endogenous GH in GH-deficient or age-related contexts1727.

Mechanism

While Epitalon works as a synthetic tetrapeptide, Ala-Glu-Asp-Gly (AEDG), that has been shown to bind histones and modulate gene expression and chromatin state, with reported effects on pineal function, neurogenesis, and longevity in animal models36, Sermorelin is a synthetic 29-amino-acid peptide identical to the 1–29 N-terminal fragment of human GHRH that stimulates GH synthesis and secretion by binding pituitary GHRH receptors1727120.

Receptor

Epitalon

Binds nuclear histone proteins (H1, H2B, H3, H4) rather than a classic membrane receptor36

Sermorelin

Growth hormone–releasing hormone receptor (GHRHR) on pituitary somatotrophs94103120

Organism or Origin

Epitalon

Synthetic peptide modeled after a pineal gland peptide fraction; not directly encoded as a standalone peptide in humans36

Sermorelin

Synthetic peptide corresponding to human hypothalamic GHRH(1–29) 17120

Summary

Epitalon and Sermorelin are noticeably different, with limited direct overlap in their usual biological context. Both are often discussed in Aging and longevity contexts, while Epitalon is more of a neuropeptide and Signal peptide and Sermorelin is better described as a hormone peptide. Their biological logic is quite different: Epitalon is a signaling modulator and a gene expression modulator, whereas Sermorelin is a receptor agonist. Both are synthetic in origin and their development context also differs, with Epitalon approved for Research use only while Sermorelin is approved. Sermorelin incorporates amidation features that are not part of Epitalon.
