DSIP vs SS-31

Function

While DSIP was originally described for inducing spindle and delta sleep activity when infused into the brain of rabbits; later studies report stress-modulating and endocrine effects, though its physiological role is still debated47, SS-31 improves mitochondrial bioenergetics, reduces oxidative stress, and shows protective effects in models of ischemia-reperfusion, neurodegeneration, and other mitochondrial disorders4637137.

Mechanism

While DSIP works as an endogenous nonapeptide, Trp-Ala-Gly-Gly-Asp-Ala-Ser-Gly-Glu, with proposed neuromodulatory actions; it interacts with components of the MAPK cascade and may be related to glucocorticoid-induced leucine zipper (GILZ), but its precise biosynthetic origin remains unknown4757, SS-31 is a mitochondria-targeted tetrapeptide, d-Arg-dimethylTyr-Lys-Phe-NH2, that selectively binds cardiolipin in the inner mitochondrial membrane, stabilizing cristae structure, improving electron transport, and reducing ROS production4637137143.

Receptor

DSIP

Not clearly established in the current dataset.

SS-31

Cardiolipin-rich inner mitochondrial membrane; interacts with cardiolipin and associated mitochondrial proteins rather than a classical cell-surface receptor46137143

Organism or Origin

DSIP

Endogenous vertebrate neuropeptide; its encoding gene has not been identified47

SS-31

Fully synthetic Szeto–Schiller tetrapeptide; not encoded in the genome4637

Summary

DSIP and SS-31 are noticeably different, with limited direct overlap in their usual biological context. Their typical research and application settings separate fairly clearly: DSIP is more often discussed in the realm of Neurology and brain health, whereas SS-31 is more often associated with the realm of Aging and longevity and Cardiovascular health. Their biological logic is quite different: DSIP is a neurotransmitter modulator, whereas SS-31 is a signaling modulator. DSIP has a more natural endogenous origin, while SS-31 is closer to synthetic design background with DSIP approved for Research use only and SS-31 in Clinical phase 2. SS-31 incorporates d-amino acid substitution and amidation features that are not part of DSIP.
