Comparison

FOXO4-DRI vs Snap-8

Function

While FOXO4-DRI acts as a senolytic in preclinical models by promoting apoptosis of senescent cells, improving tissue function and healthspan measures in aged animals38, Snap-8 is a cosmetic peptide studied for reducing wrinkle depth by inhibiting synaptic vesicle fusion and subsequent muscle contraction in facial expression muscles5060.

Mechanism

While FOXO4-DRI works as a D-retro-inverso peptide derived from a FOXO4 region that competes with endogenous FOXO4 for binding to p53, thereby releasing p53 to trigger apoptosis selectively in senescent cells38, Snap-8 is acetyl octapeptide-3 (Ac-Glu-Glu-Met-Gln-Arg-Arg-Ala-Asp-NH2), an extended analog of Argireline derived from the N-terminal region of SNAP-25 that competes with native SNAP-25 for binding to syntaxin-1, forming a non-functional SNARE complex and reducing neurotransmitter release5060.

Length and Sequence

FOXO4-DRI is amino acids long, whereas Snap-8 is longer as it has a length of 8 amino acids. FOXO4-DRI is made up of a sequence of sequence data not available in the current dataset. Snap-8 is made up of a sequence of Glutamic acid, Glutamic acid, Methionine, Glutamine, Arginine, Arginine, Alanine, Aspartic acid.

Receptor

FOXO4-DRI

Intracellular target is the FOXO4–p53 protein–protein interaction interface; it does not signal via a traditional cell-surface receptor38

Snap-8

Targets the SNARE complex (SNAP-25–syntaxin interaction) rather than a cell-surface receptor5060
