Comparison

FOXO4-DRI vs GHRP-2

Function

While FOXO4-DRI acts as a senolytic in preclinical models by promoting apoptosis of senescent cells, improving tissue function and healthspan measures in aged animals38, GHRP-2 is used in research to induce robust GH pulses and study anabolic and metabolic effects; clinically developed as pralmorelin for GH deficiency diagnostics6272.

Mechanism

While FOXO4-DRI works as a D-retro-inverso peptide derived from a FOXO4 region that competes with endogenous FOXO4 for binding to p53, thereby releasing p53 to trigger apoptosis selectively in senescent cells38, GHRP-2 is a second-generation hexapeptide growth hormone–releasing peptide that mimics ghrelin by agonizing GHSR-1a, strongly stimulating pulsatile GH release and modestly raising prolactin, ACTH, and cortisol62723096.

Summary

FOXO4-DRI and GHRP-2 are noticeably different, with limited direct overlap in their usual biological context. Their typical research and application settings separate fairly clearly: FOXO4-DRI is more often discussed in the realm of Aging and longevity, whereas GHRP-2 is more often associated with the realm of Metabolic and endocrine and Musculoskeletal health. Their biological logic is quite different: FOXO4-DRI is a protein interaction inhibitor, whereas GHRP-2 is a receptor agonist. Both are synthetic in origin with FOXO4-DRI in Preclinical development and GHRP-2 in Clinical phase 1. FOXO4-DRI carries retro-inverso isomerization features, while GHRP-2 instead reflects d-amino acid substitution and amidation changes.
