Comparison

Epitalon vs MOTS-c

Function

While Epitalon is reported to increase lifespan and reduce tumor incidence in animals, and to influence differentiation of neural and mesenchymal stem cells; is studied as an epigenetic regulatory peptide36, MOTS-c improves insulin sensitivity, enhances glycolysis, reduces oxidative stress, and shows protective effects in models of metabolic syndrome, aging, and ischemia-reperfusion injury8135146.

Mechanism

While Epitalon works as a synthetic tetrapeptide, Ala-Glu-Asp-Gly (AEDG), that has been shown to bind histones and modulate gene expression and chromatin state, with reported effects on pineal function, neurogenesis, and longevity in animal models36, MOTS-c is a 16-amino-acid mitochondrial-derived peptide that translocates to the nucleus under metabolic stress and regulates glucose and lipid metabolism largely via activation of AMPK and modulation of mTOR and folate-cycle–linked pathways854140146.

Length and Sequence

Epitalon is 4 amino acids long, whereas MOTS-c is longer as it has a length of 16 amino acids. Epitalon is made up of a sequence of Alanine, Glutamic acid, Aspartic acid, Glycine. MOTS-c is made up of a sequence of Methionine, Arginine, Tryptophan, Glutamine, Glutamic acid, Methionine, Glycine, Tyrosine, Isoleucine, Phenylalanine, Tyrosine, Proline, Arginine, Lysine, Leucine, Arginine.

Receptor

Epitalon

Binds nuclear histone proteins (H1, H2B, H3, H4) rather than a classic membrane receptor36

MOTS-c

No dedicated cell-surface receptor has been definitively identified; signaling is primarily described via intracellular AMPK and related metabolic pathways8135146

Organism or Origin

Epitalon

Synthetic peptide modeled after a pineal gland peptide fraction; not directly encoded as a standalone peptide in humans36
